Maybe I´m a little bit dumb, but: How do I use enchanted items?
And: What effects do better soulgems have?

It depends; many weapons cast their effect upon hitting an opponent, whereas rings/armor/etc. usually (especially at lower levels) have to be triggered off like a spell. Check for 'em in the same window your spells are at, and cast 'em in the same way.
"What effect do better soulgems have?"
The bigger the gem, the bigger the soul it can hold; you can catch a rat in the tiniest gem, but to capture the bigger NPCs you have to have better quality gems.

well, I got an enchanted blade, but it doesn´t seem to do anything on hit.

I have a sword that has a shield type spell built into it. You need to select it like an amulet, then cast it like a spell before you get the effect. When you look at an weapon you'll see one of three phrases:
cast upon use = use like an amulet
constant effect = always effective
cast upon strike = hit a creature and the spell is cast

If the item has a "Cast on Use" enchantment, you have to manually activate it. When you right-click to look at your inventory, character screen, map, etc.. there's also a magic menu that pops up. On it are listed: any spells you know, any magic items you are carrying, and any scrolls you are carrying. Selecting an item from the list will ready it for casting. When you right click again to go back to playing, press 'R' to go into magic using mode. Then just click the left mouse button to cast the spell.
