I would very strongly recommend playing it single-player first.
You can never get the full feeling of SS2's excellent storyline when you play it multiplayer.

I would definitely recommend playing the entire game through at least once in singleplayer before even thinking about trying multiplayer.
You will get a much better experience of the plot.
Nothing ruins immersion more than some fool with a wrench jumping up and down and attacking the walls during cutscenes...
