They frob without getting highlighted. All you need to do is to face them and press your "frob" key.

How do I know if they're frobbed if they don't highlight or make some kind of noise?

You only have to
get close to them and use them. The problem is that you didn't find the three glyphs, one is near Garret's House, the second is near a door in the streets and the third is in the third floor of the Pub.
