Of course, this may never get off the ground but it's a pretty awesome concept that you might one day send off a group of humans who will never once return to Earth but valiantly stride out to colonise new worlds... and new civilisations... boldly going... etc
The bit about genome manipulation sounds like a bit of a red herring though.
"We're thinking about colonising Mars... oh, but we'll probably alter the human genome to allow for people to breathe carbon dioxide first."

Wouldn't radiation be a major issue? Is it even theoretically possible to have some material that could block it?

From what I've read a few feet of martian rock and regolith would be enough. Well i don't know exactly how much, but living underground at least should be an option.
