Nope.
Neither is necessary. You just have to use the vent shafts to go to the other side of the station and then wait until the NSF is gone on Patrol and talk to the hostages while staying hidden behind the pillar
Usually I even go there before going for the Ambrosia. First, Anna loves the surprise, and second, she gives you the EMP grenades anyway. ;)

I have rescued the hostages before I went for Ambrosia, but Navarre has discovered the container first.

The trick is to not approach Castle Clinton, or the soldiers will open fire, thus leading to the possible discovery of the Ambrosia by Anna.
(It never happened to me though. It's the first time I hear Anna can discover that canister...)
Unless of course you went in the subway with the hostages. You have to crawl back to the vent shaft to finish the job at CC. If you go downtown and come back, it's over, I think.

Anna can detect the canister with ambrosia? Wow, I wouldn't have thought... :)
A nice swing for the Subway Station (if you are annoyed or just want to play it in the gory way) is to let the hostages run into the train, wait until the door of the train is closed shut again, and then blow up the whole station by shooting at one of the crates of boom. :ebil:
But well, you don't need to do something gory like this, is it indeed possible to sneak in and out without getting spotted by anyone of them.

That freaking training mission. Went through the whole thing twice just to pick up the TNT toss it at the security bot and get my legs blown off.
I beat the game once... maybe I'll go back and show that stupid training mission a thing or two.
