I'm on part of the main quest where I have to go to that sea cave near Gnaar Mok. I've managed to kill that main priest without getting hit once, but when I check my stats next I always end up with corprus. Is this a result of reading the letter that Dagoth sent me? If so, do I have to read it to progress in the game?

It's part of the plotline.
Get back to Caius and he'll get you on the right track.
If I remember right, it's a result of Dagoth Gares cursing you. You should get a message window that pops up as you kill him.

If you mean Khartag Point as "the huge boulder", try crossing the water to the south; the 1st land you run into is bordered with neck-high rocks, and the entrance is just behind them.

I killed Dagoth Ur without getting Corprus, Wraithguard, named Nevarine or Hortator...cuz I goofed up.
I ran around too long leveling up, and the Orcish mage in Balmora disappeared, leaving me stranded in the main quest.
I killed the ultimate baddy (it was tough, sunder almost did me in!), and nobody seems to care...:(.
