Hi,
OK so the snakewomen told me about the portals and give me the secret magic
combination mega spacium to activate the portals.
But : How do you activate these portals ? The rune combination is not in my spellbook.
What is the combination and can I use it this early in the game ?

IIRC, she also gives you the runes... Perhaps check your inventory, or look on the ground where she talked to you?

I was confused by this too, because the "spell" is not listed in your book (like others where you can click on them for easy reference), but you do have the runes and you can still cast the spell. Just click on the mega & spacium runes if you need help with how to cast them.

I'm having difficulty with this too , I got the two runes from the snake woman , and I thought they would allow me to use the portals {rather than all that frankly tedious upping and downing staircases }. But I gather it's just for getting through the Snakewomens' door . So how do I use the portal ? I'm fairly advanced into the game but maybe I'm missing a few runes ? { so to speak }:confused:

AFAIK the mega spacium spell only activates portals (the sisters' door being opened is only a matter of them deciding that it's time to let you in).
One activated portal won't do anything until a second one has been activated. Only then you'll have a destination to teleport to: The other portal's location image will appear on one of the sides of the portal, and you should be able to double click this, thus starting the teleportation.
Each additional portal that is activated, should appear in all the other activated portals as a possible destination.
Do note, that for the activation of a portal, you have to stand right next to it when you cast the spell.
