I've heard there is a secret way into the Sisters Hideout and a treasure room to be looted in there.
How do I get in?

There is a hidden entry on level four through an entrance that you can find if you climb some of the disjointed scaffolding.

Just don't get there before you're supposed to; it'll turn the Sisters permanently hostile and you'll need them for a mission.
EDIT: This happened to me early this year, and I am now awaiting the decay of my memory of the game so I can replay it freshly again.

I made that mistake. I managed to get in with a spawn undead spell. I spawned a zombie past the barrier and when it started to attack the snake bitch, the barrier opened and she came after me. I was able to run inside after that. I've recently started playing it over again.

Brilliant! Thank you. I never thought to look at scaffolding, I'd seen it marked on my map and thought maybe I need a key or something and a higher skill to see where the entrance was.

I'd like to know the answer to this as well. I also went there a couple of times on quests and so on, but don't know what is meant by the Treasure Room? Did I miss out on some loot?
